A Box-Behnken design for optimal green extraction of compounds from olive leaves that potentially activate the AMPK pathway

Olive leaves hide a secret weapon against obesity—and scientists just found the perfect green recipe to unlock it.

For centuries, olive leaves have been more than just a symbol of peace—they're a treasure trove of bioactive compounds. Among these, certain molecules can activate AMPK, a key enzyme that helps reduce lipid accumulation inside cells. That makes olive leaves a promising candidate in the fight against obesity. But how do we extract these compounds efficiently without harming the environment? Enter microwave-assisted extraction (MAE), a green technique that uses energy-efficient microwaves to pull out phenolic compounds from plants. In this study, researchers used a Box-Behnken design to fine-tune three MAE parameters: temperature, ethanol concentration, and extraction time. They aimed to maximize both the total compound yield and the amount of AMPK-activating compounds. After analyzing every extract with high-performance liquid chromatography coupled to mass spectrometry, they found two distinct optimal sets of conditions. To balance both goals, they applied a desirability function, landing on 111°C, 100% ethanol, and 23 minutes—with a desirability score of 0.97, indicating near-perfect outcomes. The resulting olive leaf extract holds great potential for obesity treatment, offering a green, effective path forward.
