What if the key to fighting liver cancer lies in a simple water extract? A new study reveals that water, not ethanol, may unlock powerful antioxidants and anti-cancer activity from a natural source.

Forget complicated chemical cocktails—sometimes nature's simplest solvent holds the most power. In a recent study, scientists optimized the extraction of phenolic compounds from a plant source (TSH) using a 2-level factorial design, testing 60% ethanol and distilled water at different temperatures. The goal? To maximize total phenolic content (TPC) and evaluate antioxidant and anti-liver cancer activities.

The results were surprising. While the ethanolic extract boasted higher TPC, the water extract stole the show. It exhibited stronger antioxidant activity and dramatically more potent cytotoxicity against HepG2 liver cancer cells, with an IC50 of 44.1 µg/mL after 72 hours—compared to 408.3 µg/mL for the ethanol extract. This effect was time- and dose-dependent, and the water extract also inhibited cell migration, a key step in cancer spread.

Advanced analyses (HPLC and LC-ESI-QTOF-MS/MS) identified oligomeric proanthocyanidins in the extracts, hinting at the bioactive compounds behind these effects. The study's optimal model equation (TPC = 0.9 - 0.0064A + 0.0059B + (2.61 × 10-5)AB) offers a predictive tool for future extraction optimization.

This research underscores the potential of water as a green, effective solvent for extracting health-promoting compounds. The water extract of TSH could become a promising candidate for functional foods and natural anti-liver cancer therapies—proving that sometimes, the simplest approach is the most powerful.
