Effect of extraction methods on yield, phytochemical constituents and antioxidant activity of Withania somnifera

How you brew your herbal medicine matters just as much as the plant itself. When it comes to the revered root of Withania somnifera, the secret to unlocking its medicinal power lies in the extraction technique.

Withania somnifera, a celebrated wonder herb deeply rooted in Ayurvedic medicine, packs a powerful punch of active compounds. Its roots are famous for withanolides—distinctive steroidal lactones acting as key chemical markers—alongside beneficial phenolics and flavonoids. Traditionally, traditional herbal preparations rely on simple water extraction through infusion or decoction.

To see if we can do better, researchers put modern "Green Extraction" techniques to the test. They compared conventional extraction methods against cutting-edge ultrasound-assisted and microwave-assisted solvent extraction, using both water and water-alcohol solvent systems.

The results reveal that the extract yield, the concentration of total phenols and withanolides, and the resulting antioxidant activity—measured via DPPH and ABTS assays—are not static. Instead, they shift dramatically depending on both the chosen extraction process and the precise solvent composition used.
