Natural Deep Eutectic Solvents Enhance the Bioavailability and Antioxidant Activity of Oleanolic Acid in Self-Constructed Pickering High Internal Phase Emulsions

Imagine a natural compound so powerful yet so poorly absorbed that most of it goes to waste. Now, scientists have found a clever way to trick the gut into taking it up—using nature's own solvents.

Oleanolic acid, a plant compound with promising antioxidant powers, has a problem: the body barely absorbs it. But a new study turns that around with a clever trick—wrapping it in a special emulsion where natural deep eutectic solvents (NADESs) replace water as the inner phase. These solvents, made from simple molecules like proline and glucose, form a hydrogen-bonding network that stabilizes the compound and boosts its solubility.

In lab tests simulating digestion and intestinal absorption, the NADES-based emulsions dramatically improved oleanolic acid's bioaccessibility. The bioavailability jumped to 16.20–19.10%, far surpassing the control group's levels. The secret lies in the hydrogen bonds, which seem to help the compound slip through intestinal cells more easily.

The benefits didn't stop there. When cells were stressed with an oxidant, the emulsions significantly reduced damage—lowering harmful MDA and ROS while boosting protective enzymes like SOD and CAT. Interestingly, the NADESs themselves were major contributors to this antioxidant effect.

This approach offers a simpler, more stable delivery system for hydrophobic phytochemicals. By using nature's own solvents, we might finally unlock the full potential of compounds like oleanolic acid—turning a good molecule into a great one.
