Pulsed Electric Field-Assisted “Green” Extraction of Betalains and Phenolic Compounds from Opuntia stricta var. dillenii Prickly Pears: Process Optimization and Biological Activity of Green Extracts

What if a simple electric zap could make fruit extracts healthier and greener? Scientists just found a way to boost antioxidant yields from prickly pears by up to 135%—using nothing but water and a pulse of electricity.

Prickly pears are not just desert survivors—they're packed with betalains and phenolic compounds, natural molecules linked to health benefits. But extracting these treasures usually requires harsh solvents and heat. Now, researchers have turned to a gentler ally: pulsed electric fields (PEF).

In this study, whole Opuntia stricta var. dillenii fruits were pre-treated with brief electric pulses to break open cell walls, then soaked in plain water. Using a statistical design, the team optimized the process: a 10.5 kJ/kg pulse at 5 kV/cm, followed by a 35°C water bath for 165 minutes. The result? Compared to a conventional ethanol-based extraction, the PEF-assisted method boosted total betalain yields by 61% and phenolic compounds by 135%.

The benefits didn't stop at quantity. Antioxidant activity jumped—DPPH scavenging rose 145%, FRAP increased 28%—and anti-inflammatory potential improved by 19%, as shown by hyaluronidase inhibition. High-performance liquid chromatography confirmed the presence of diverse bioactive compounds.

This approach proves that green chemistry can be both effective and sustainable. By using water as the only solvent and a simple electric pre-treatment, we can unlock more of nature's pharmacy without leaving a heavy environmental footprint.
