RP-HPLC-PDA-ESI-MS/MS screening of bioactive compounds from Degla-Beida dates: Conventional and green extraction technologies

Dates aren't just a sweet treat—they're a powerhouse of bioactive compounds with therapeutic potential. In this study, scientists set out to identify these compounds in Degla-Beida dates, using a sophisticated technique called RP-HPLC-PDA-ESI-MS/MS. They compared three extraction methods: microwave-assisted (MAE), ultrasound-assisted (UAE), and conventional extraction (CE), to find the most efficient way to pull out the phenolics.
The team used response surface methodology (RSM) to optimize the microwave extraction. Their model showed that a quadratic polynomial best described the total phenolic content. The winning conditions? A 50% ethanol concentration, 700 W microwave power, and just 2.42 minutes of extraction time—yielding a maximum TPC of 4.27±0.09 mg GAE/g DW.
HPLC analysis revealed a diverse array of compounds: organic sugars and acids, nucleosides, carotenoids, proanthocyanidins, lignans, flavonols, flavones, saponins, and sterol glucosides. The optimized MAE extract also showed strong antioxidant activity, with 58.72±0.29% DPPH radical scavenging and a reducing power of 1.88±0.09 mg AscAE/mL.
This work highlights how green extraction technologies can efficiently unlock the health-promoting compounds in dates, offering a sustainable path for nutraceutical development.
