The effect of ultrasonication pretreatment on the production of polyphenol-enriched extracts from Moringa oleifera L. (drumstick tree) using a novel bio-based deep eutectic solvent

Could a 30-minute ultrasound bath be the secret to unlocking more antioxidants from moringa leaves? Or is it just a noisy detour?

Moringa leaves are a treasure chest of polyphenols—natural compounds with impressive bioactivity. But extracting them sustainably is a challenge. Enter a novel green solvent made from glycerol and nicotinamide, a natural deep eutectic solvent (NADES) that promises eco-friendly extraction.

In this study, researchers tested whether zapping the leaves with ultrasound before a standard stirred-tank extraction could boost the yield. Using response surface methodology, they found the maximum total polyphenol yield—82.87 mg gallic acid equivalents per gram of dry mass—after 30 minutes of ultrasonication. But here's the twist: that yield wasn't significantly different from the non-treated samples.

Digging into the kinetics, the pretreated samples demanded more activation energy, likely because the ultrasound washed away the easy-to-extract polyphenols early on. Chemical analysis confirmed the polyphenolic profiles were identical, with only a hint of improved extractability from the pretreatment.

So, is ultrasonication worth it? The data says: not really—but it doesn't hurt. For green extraction, the solvent is the star; the ultrasound is just a supporting actor.
