The Protective Anticancer Effect of Natural Lycopene Supercritical CO2 Watermelon Extracts in Adenocarcinoma Lung Cancer Cells

What if a natural compound in watermelon could fight lung cancer cells while synthetic versions fall short? A new study reveals that the source of lycopene matters—and the results are surprising.

Lycopene, the pigment that gives tomatoes and watermelons their red hue, has long been hailed as a cancer-fighting antioxidant. But not all lycopene is created equal. In a recent study, scientists tested three natural oleoresins—extracted from watermelon, gấc, and tomato using green supercritical CO2 technology—against synthetic lycopene on human lung adenocarcinoma cells (A549). The results were anything but uniform.

At the same concentration (10 µM), the watermelon extract (Lyc W) stood out dramatically. It increased apoptosis, mitochondrial stress, and reactive oxygen species (ROS), while also boosting NF-kB and IL-8 expression—and significantly reduced cell proliferation. In contrast, the gấc and tomato extracts (Lyc G and Lyc T) only increased cell proliferation, and synthetic lycopene had no effect at all.

The key difference? Lyc W had the highest lycopene-to-tocochromanols ratio. This suggests that the balance of compounds, not just lycopene alone, determines the outcome. The study warns that antioxidant tocochromanol supplements might not only lack anticancer benefits but could even enhance cancer aggressiveness—unless lycopene is bioavailable enough to counteract them.
