Ultrasound-assisted extraction and flavor quality assessment of in vitro biomimetically fermented Kopi Luwak

What if the world's most controversial coffee could be made without a single civet involved? Scientists have now brewed a Kopi Luwak alternative in the lab that out-scores the original.

Kopi Luwak's legendary flavor comes at a steep price—both ethical and environmental. But a new study has cracked the code by recreating the civet's digestive magic in a flask. Using metagenomic data from the civet gut, researchers selected 30 core functional strains from 1,870 isolates to build a synthetic symbiotic consortium. This microbial team, optimized through a three-stage process involving single-factor experiments, Box–Behnken design, and a GA-ANN model, faithfully mimics nature's fermentation.

The winning recipe—16.5% inoculum, pH 6.25, 33°C, and 135 hours—delivered an average SCA cupping score of 82.92, with a top score of 85.25. That's significantly higher than both natural fermentation and the real thing. The lab-grown brew also boosted total acidity to 0.78 g/L and polyphenols to 225.3 mg/L, while increasing key flavor compounds like 2,3-dimethoxyphenol by up to 2.3-fold. Bitter culprits like caffeine dropped by 41%.

Ultrasound-assisted extraction proved the perfect partner, recovering roughly 28% more volatiles than conventional methods while slashing energy use by at least 66% and halving solvent needs. The result? A cleaner, greener, and arguably tastier cup of coffee—no civets harmed.
